GENETIC CODE of  6 LETTERS

Floyd Romesberg  (Scripps Research, La Jolla, Ca.) et al,  inserted  two  new  nucleotides  in vitro: d5SICS-DNAM,  into the DNA of  the  bacterium  E. coli,  expecting   the new sequence   expand the genetic   code,  increase  its potential,  produce new proteins (by having more than  20 amino acids), get  new therapies  and  create    new tasks (possibility of replacing the current code). Cellular enzymes copied the 2 letters with high efficiency and fidelity  and  the resulting  bacteria propagate  in  stably form, the  expanded genetic alphabet. Romesberg  says  that  if any bacteria escape  from  the laboratory  to the  environment, this would revert to  his DNA of 4 letters. When   we   observe  our genetic code and   perceive the existence of 4 letters (nucleotides) and 2 pairs (GC and AT), we  ask ourselves : Why 4 letters? Why not six? 8? or 10? The   mathematician  E. Szathmary hypothesized, if the alphabet were larger, letters begin to resemble each other fairly, causing  base pairs mispairings and  a greater number of mutations. Biologists, like:  C. Woese and others argue that the 4 letters and  its  combinations reached an optimal,  paradoxically non-evolutionary state (frozen), so that  nucleic acids stored and express genetic information in the form of enzymatically active RNA molecules (ribozymes), allowing copying   general information with fidelity, self-reproduction and creation of efficient metabolic pathways. The current genetic code also exhibits ability to change, a  basic feature to evolve.

FIGHT  AGAINST    AGING
I)In recent years have  been created   biopharmaceutical companies, decided to  understand  aging,  in order  to  modify aging processes  and  if possible extend  the life. According to an article published in Nature Medicine, the company Calico, with  stars  researchers  like : A. Levinson, HV Barron, D. Botstein, C. Kenyon (discoverer of nematode genes that lengthen  their  lives), RL Cohen and Lewis JW, work   for    this purpose. Together with the company AbbVie, Calico, plans to invest $ 1.5 billion in research and development of anti-aging  medicines. Another company: Human Longeviy, led by Craig Venter, focuses his studies directly in humans, hoping to sequence the genes of 100,000 people per year, to analyze their protein profiles, microbial content  of  their bodies and  digitalize images  of their  bodies. Some drugs discovered are:  P7C3 compounds, resveratrol, metformin, sirtuins.  II) For us, the success of these companies must be based on updated, human holistic conceptions of   growth-aging  systems. Let's see: a) We know that  first mammals (of whom, we descend), evolved from reptiles, who unlike the former, do not increase their mortality with age, regenerate oocytes  and limbs and continually replace their teeth.  Some  researchers think  that  due to  the smallest size of  primitive mammals, these adopted the evolutionary strategy to survive and reproduce  early and  aging  rapidly.  So  the greatest contribution of natural selection  to  breed new generations would come mainly from young mammals or people. For these reasons and according to Peter Medawar (1952), the positive force of natural  selection decreases with age. The recent discovery of GDF11  protein, in the blood of young  mice, induced by protective genes, appears to  confirm   this  theory. According to William´s  model, some beneficial genes in childhood would become dangerous (accumulation of mutations), during aging. b) Kirkwood and Austad (2000), argue  that aging occurs due to accumulation of cellular damage  and energy loss  induced by  multiple  wear and defensive-reparative  mechanisms employed throughout our lives. c) As  mentioned  before,  several  companies are developing  antiaging specific medicines (targeted), usually created by genetic engineering. These specific drugs are  focused by the time to  the most prevalent diseases in aging people: cancer, diabetes and neurodegenerative disorders. d) The `problem  is that   biopharmaceutical companies will not investigate and  treat  the fundamental    problem of aging,  the same  that  according  to  L. Hayflick,  occurs after   mammals  reach  reproductive maturity,  when  a thermodynamic instability  is initiated,  breaking the  somatic machinery (genome),  in charge  of maintaining the order and  molecular effectiveness  (repair and replacement of cells),  urging  to Hayflick  to make the fundamental question in aging  biology  ¿what changes in biomolecules  lead  to increased vulnerability and functional decline of the body? e) An ingenious anti-aging proposal  suggested by Haldane is : postpone sexual maturation  with drugs as much as possible (at 120 years,  we would have bodies of children).

ANTI-AGING EFFECT  in ADULT MICE

The recent discovery of anti-aging effects in adult mice after blood transfusions coming from young mice  -from our point of view-  is solid and  have  human perspective, because: a) One of five,   identified antiaging protein : growth differentiation factor 11 (GDF11), in the serum  of  young  donor mice is similar to that in humans. b) The physiology of young and senile mice is different. In young mice prevail growth promoting factors : in size and volume, as well as functional efficiency of cells (synaptic plasticity, cognitive efficiency) and better organs (best bodies). Conversely, in aged mice, the body tries to avoid wear and functional decline of the brain, heart, muscle strength, etc.). c) It would be interesting to investigate whether blood transfusion of old mice to young mice, slows growth and development of the last.

The recent launching  (16/12/2014), of the  Geostationary Satellite Launch Vehicle (GSLV), Mk- III, rocket by the space agency : ISRO, places  Hindus to the level of  : Chinese, American European and Russian  space agencies, because: I) The GSLV is a rocket of 630 tons, for terrestrial and extraterrestrial long range missions (interplanetary and multi-satellite launches).  Let's look its  unmanned capsule (designed to accommodate 3 human crew),  of 3735 kg , which was separated and landed successfully in the Indian ocean, 20 minutes after departure, destined to descend on the Moon by 2017 and by 2018, on Mars. II) The GSLV employs a powerful cryogenic engine, continuously upgraded. III) Show the world the existence of a successful upscale techno-scientific Hindu knowledge. IV) The synthesis of this human group  focused on priorities (without neglecting the practical), achieve successful launches with limited financial budgets. The cost of the recent launch of GSVL, was only $ 25 million. V) To make matters better, India is beginning to reap profits after entering to a market of $ 300 billion, advising and assisting developed countries like:  Germany, France, Canada, etc in space and related fields. 3 years ago,  Hindus discovered in the upper atmosphere (height : 27-30 km), 3 species of extremophile bacteria (one called: Bacillus isronensis), extremely resistant to ultraviolet radiation.

OPTOGENETICS

Thinking  about   the difficulties and slow progress in brain research, Crick (1999. 354: 221-25), wrote: "a)...When we  understand scientifically our perceptions, thoughts and emotions, the vision of ourselves and the universe will be totally transformed ...b) ... After identifying the different types  of neurons, the next big step will be  to put one or more neurons quickly, in on-off, using infrared light ... in a particular light sensitive cell”. If we wanted to scrutinize today, one of the avenues through which will pass  techno-science very fast in the coming years, the selected would be, no doubt : optogenetics. Some of   leading researchers in this area: Karl Deisseroth, Edward S. Boyden, Feng Zhang, Peter Hegemann, Gero Miesenböck, Georg Nagel, etc., may soon receive a Nobel Prize. Boyden (MIT), says that his  optogenetic investigations are possible because the brain (organ, about which many  thought we would never understand its operation), works  with binary codes that can be explored and understood, easily. If so, it would be possible for us to be bio-robots with binary codes programmed by nature or someone. Therefore,  artificial intelligence with programmable codes and emerging roles  should reach the same position and sophistication of modern human brains.

Current experiments in optogenetics, excite (on, gain of function) or inhibit (off, loss of function), brain cells of experimental animals in vivo, shooting rays of light rays  to  neuronal surfaces, previously sensitized with rhodopsin (protein sensitive to visible light), extracted  from environmental microorganisms, allowing researchers  to perform accurate temporary registers with optogenetic sensors for calcium, chlorides, voltages membranes, etc. Preconditioning of neurons is performed  translating opsin-genes (via  virus carriers), extracted from  envirommental, unicellular  fluorescent algae to  neurons of   experimental animals. Using channelrhodopsins to excite  neurons or silencing them with halorhodopsins,  allow us  a  better understanding about how normal and abnormal neural circuits, functions. It is  expected soon to know the causes of certain neurological diseases (Parkinson disease, autism, schizophrenia, drug addiction, anxiety, depression, etc.), to treat them properly. It is also expected to control inappropriate behavior of animals and why not of certain humans.

OPTOGENETICA

AN OCEAN at ENCELADUS

WATCH  VIDEO

Most scientists suspected that water existed on Mars since ancient times, judging by its meandering channels. Today, the almost certainly discovery of liquid water beneath the icy south pole surface of Enceladus (one of the 62 moons of Saturn), could have unpredictable connotations : extraterrestrial life (Archaea genus), migrate and colonize the outer space, possibility that human life has an extraterrestrial origin (panspermia). According to David Stevenson (California Institute of Technology/Pasadena), this ocean could extend from pole to pole, contacting useful rocks for life: phosphorus, sulfur, potassium. The Cassini spacecraft photographed the 2005 steam flows expelled through fissures of the south pole of Enceladus, motivating to Nikolai Brilliantov (Leicester University), to be the first to argue that such flow of water vapor indicated the existence of an ocean submerged in Enceladus, inducing several studies recently published in Science.

Luciano Sapienza (Rome University) and colleagues deduced the existence of the ocean, using measurements taken during 3 fly- bys (April 2010-May 2012), when the probe flew 100 km height over Enceladus. Using a network of giant antennas of NASA, researchers monitored radio signals from the probe, looking for subtle changes in their frequencies, revealing that the probe accelerated few mm/sec, up or down. Some of these changes were due to speed variations of the gravitational field of the moon, induced by the different densities of material located beneath its surface. The researchers mapped the gravitational field of Enceladus: denser than ice, less dense than rocks located beneath its south pole, allowing Stevenson to sustain that heated ice becomes liquid water thanks to gravitational forces (tidal heating), acting on Enceladus as it orbits Saturn. It is suspected that this ocean was initially heated with circulating water contacting solid silicate, which could have allowed life, due to the antifreeze effect of salt : reducing the temperature. Today, it is planned to send a probe to Enceladus to analyze the content of water vapor and to search for biomolecular evidence of life.

BLACK HOLES ARE STORES ...

A radical proposal of Stephen Hawking, regarding the event horizon of a black hole (until recently defined as a closed area of space from which not even light, could escape), attempts to emphasize known notions of space-time or perhaps : modify them. The new proposal of Hawking argues that: 1- There are no rigid horizon events...light and matter could escape from these holes. According to Hawking, quantum effects around the black hole (high energy region), would make space-time fluctuate there wildly, within very narrow limits. 2- Instead of an event horizon, there would be an apparent horizon, an area where many rays of light would have suspended, trying to escape from the core of the black hole. General relativity has allowed some physicists postulate an explanatory model in this regard: if more material is swallowed by the black hole, the event horizon will grow more than the apparent horizon. If the black hole expels radiation, the event horizon will be smaller. Matter and energy, would only temporarily maintained after the apparent horizon, being compressed like an accordion without reaching the core, just to try to escape again and again. That nothing can escape the event horizon is for Hawking incompatible with quantum theory. An apparent horizon could be dissolved when compression of matter subjected to mechanical and quantum-gravity effects, reaches certain limits. As Hawking argues that apparent horizon is the actual limit and that its eventual dissolution could permit the escape from anything contained in a black hole, some physicists argue that such proposal would mean absence of singularities and black holes. At the end, it seems that black holes are flexible matter-energy stores, to maintain normal functioning (fine tuning),of the universe.

BREAKTHROUGH : MONKEYS GENETICALLY MODIFIED

BREAKTHROUGH : MONKEYS GENETICALLY MODIFIED As we foresaw in previous articles, techno-scientific changes in the next 10 years will take fancy characters in certain cases and many of them will happen very faster. A couple of monkeys (Long tail macaques Cynomolgus), genetically modified, created via cut and paste DNA, are proof of this. The study of these twins monkeys created in Nanjing Medical University-China, is published in Cell. They are the first primates having an altered genetic pattern. The next step will be -no doubt- to create new human genomes (that will provide their holders greater physical strength, more longevity and absolute resistance to cancers and other diseases). To do this, Chinese researchers led by Jiahao Sha, used a system developed in USA in 2005: CRISPR/Cas9, an editing gene system (to mute, enhance or change specific genes), in different organisms and target precise monkey genome points to manipulate 2 genes (coinjection of mRNA and sgRNAs Cas9 in unicellular embryos, yielding a precise gene targeting in Cynomolgus monkeys fertilized eggs, before transferring it to surrogate mothers. The experiment received no critic in China because in this country there are no laws or strict ethical controls related to animal experiments. In this regard, researchers of several parts of the world argue that the method will remove defective cellular genes, replacing them with others healthy. Will allow also to recreate in monkeys : Alzheimer's and Parkinson's diseases and many people will become resistant to HIV preventing the virus from entering to human immune cells, after removing the CCR5 gene thereof. Since many years ago, many scientists advocate that experimental models based on cell and human tissues should be established to create better human responses, the chinese study attempts also to cultivate organoids (small acini of human organs), devoid of certain genes to study the cause of many diseases, analyze characteristics of drugs and monitor at acceptable costs the progress of serious diseases. If Crispr/Cas9 edition works in primate brain (similar to humans), experiments, formidable results will be obtained. Less optimistically, other researchers argue that animal rights will be violated and that eventually could be created an unlimited number of sick animals.

VITAMINS and AGEING

According to David Sinclair (New South Wales-Harvard University) and collaborators a decreased supply of nuclear reduced nicotinamide adenine dinucleotide NAD+, cause pseudohipoxia and a deteriorated communication between nuclear genes and their corresponding encoded mitochondrial genes (genomic asynchrony, with functional reduction of mitochondrial oxidative pathway genes), promoting bodily aging. The researchers showed that raising nuclear NAD+ levels in old mice prevented the seudohipoxia and metabolic dysfunction, reversing aging in just one week . Based on this experiment some companies are preparing to sell miracle pills containing: NAD+ or parts of it, in order to restore nuclear-mitochondrial communication, thus maintaining the effectiveness of multiple metabolic pathways. NAD+ is a coenzyme whose primary function is the exchange of electrons and hydrogenions during energy production in all cells. It is possible to get complete components of this coenzyme from the vitamin niacin, contained in food. If rejuvenation occur in humans, it would confirm some of the postulates of the British gerontologist Aubrey de Grey (The Mitochondrial Free Radical Theory of Aging), who accused old age as a disease likely to be cured if preventive measures (ingestion of all kind of vitamins), are implemented. Would also fulfill some of the tenets of biochemical Linus Pauling, Nobel Prize in Chemistry 1954, who since the 70s recommended that humans must ingest megadoses of all kind of vitamins, including vitamin C: ascorbic acid (Orthomolecular Medicine), useful according to him, to stop aging and cancer.
