GENETIC CODE of  6 LETTERS

Floyd Romesberg  (Scripps Research, La Jolla, Ca.) et al,  inserted  two  new  nucleotides  in vitro: d5SICS-DNAM,  into the DNA of  the  bacterium  E. coli,  expecting   the new sequence   expand the genetic   code,  increase  its potential,  produce new proteins (by having more than  20 amino acids), get  new therapies  and  create    new tasks (possibility of replacing the current code). Cellular enzymes copied the 2 letters with high efficiency and fidelity  and  the resulting  bacteria propagate  in  stably form, the  expanded genetic alphabet. Romesberg  says  that  if any bacteria escape  from  the laboratory  to the  environment, this would revert to  his DNA of 4 letters. When   we   observe  our genetic code and   perceive the existence of 4 letters (nucleotides) and 2 pairs (GC and AT), we  ask ourselves : Why 4 letters? Why not six? 8? or 10? The   mathematician  E. Szathmary hypothesized, if the alphabet were larger, letters begin to resemble each other fairly, causing  base pairs mispairings and  a greater number of mutations. Biologists, like:  C. Woese and others argue that the 4 letters and  its  combinations reached an optimal,  paradoxically non-evolutionary state (frozen), so that  nucleic acids stored and express genetic information in the form of enzymatically active RNA molecules (ribozymes), allowing copying   general information with fidelity, self-reproduction and creation of efficient metabolic pathways. The current genetic code also exhibits ability to change, a  basic feature to evolve.

Coli, esperándose  que la  nueva secuencia  expanda el código, incremente  su potencial,   produzca  nuevas proteínas (al disponer de más de 20 aminoácidos),  se creen  nuevas terapias y nuevas tareas (posibilidad de reemplazar al código actual).  Las enzimas celulares copiaron  las   2 letras con alta eficiencia y fidelidad y la bacteria resultante propago de forma estable el alfabeto genético expandido.  Romesberg,  asegura  que si alguna bacteria  escapase del   laboratorio al medio ambiente, esta  revertiría a su DNA  de 4 letras.   Cuando al observar nuestro  código genético  percibimos la existencia de 4 letras (nucleótidos) y 2 pares de bases  (G-C y A-T), nos autopreguntamos: ¿Porque 4 letras?  ¿Porque no 6? 8? o 10?  Según  hipotetiza el matemático E. Szathmary, si el alfabeto fuese más  grande,  las letras empezarían a parecerse bastante entre , ocasionando  inadecuados  emparejamientos de pares de bases   y un mayor número de  mutaciones. Biólogos de la talla de C. Woese y otros sostienen que las  4 letras y sus combinaciones alcanzaron un estado optimo,  paradojalmente  no evolutivo (helado),  para que los  ácidos nucleicos almacenaran  y  expresaran información genética en forma de moléculas de  RNA enzimáticamente activas (ribozimas), permitiendo  fidelidad del copiado de la información general, autoreproducción  y creación  de vías metabólicas eficientes. El código genético actual exhibe además capacidad de cambio, característica básica  para evolucionar.

FIGHT  AGAINST    AGING
I)In recent years have  been created   biopharmaceutical companies, decided to  understand  aging,  in order  to  modify aging processes  and  if possible extend  the life. According to an article published in Nature Medicine, the company Calico, with  stars  researchers  like : A. Levinson, HV Barron, D. Botstein, C. Kenyon (discoverer of nematode genes that lengthen  their  lives), RL Cohen and Lewis JW, work   for    this purpose. Together with the company AbbVie, Calico, plans to invest $ 1.5 billion in research and development of anti-aging  medicines. Another company: Human Longeviy, led by Craig Venter, focuses his studies directly in humans, hoping to sequence the genes of 100,000 people per year, to analyze their protein profiles, microbial content  of  their bodies and  digitalize images  of their  bodies. Some drugs discovered are:  P7C3 compounds, resveratrol, metformin, sirtuins.  II) For us, the success of these companies must be based on updated, human holistic conceptions of   growth-aging  systems. Let's see: a) We know that  first mammals (of whom, we descend), evolved from reptiles, who unlike the former, do not increase their mortality with age, regenerate oocytes  and limbs and continually replace their teeth.  Some  researchers think  that  due to  the smallest size of  primitive mammals, these adopted the evolutionary strategy to survive and reproduce  early and  aging  rapidly.  So  the greatest contribution of natural selection  to  breed new generations would come mainly from young mammals or people. For these reasons and according to Peter Medawar (1952), the positive force of natural  selection decreases with age. The recent discovery of GDF11  protein, in the blood of young  mice, induced by protective genes, appears to  confirm   this  theory. According to William´s  model, some beneficial genes in childhood would become dangerous (accumulation of mutations), during aging. b) Kirkwood and Austad (2000), argue  that aging occurs due to accumulation of cellular damage  and energy loss  induced by  multiple  wear and defensive-reparative  mechanisms employed throughout our lives. c) As  mentioned  before,  several  companies are developing  antiaging specific medicines (targeted), usually created by genetic engineering. These specific drugs are  focused by the time to  the most prevalent diseases in aging people: cancer, diabetes and neurodegenerative disorders. d) The `problem  is that   biopharmaceutical companies will not investigate and  treat  the fundamental    problem of aging,  the same  that  according  to  L. Hayflick,  occurs after   mammals  reach  reproductive maturity,  when  a thermodynamic instability  is initiated,  breaking the  somatic machinery (genome),  in charge  of maintaining the order and  molecular effectiveness  (repair and replacement of cells),  urging  to Hayflick  to make the fundamental question in aging  biology  ¿what changes in biomolecules  lead  to increased vulnerability and functional decline of the body? e) An ingenious anti-aging proposal  suggested by Haldane is : postpone sexual maturation  with drugs as much as possible (at 120 years,  we would have bodies of children). 

LA  LUCHA CONTRA  el  ENVEJECIMIENTO
I)En los últimos años se han creado compañías biofarmaceuticas, decididas a comprender  mejor  los procesos fundamentales del envejecimiento,  modificarlos  con fines  anti-envejecimiento  y si es posible   alargar la vida. Según un  artículo publicado  en Nature  Medicine,  la compañía Calico, con  investigadores  de la talla de : A. Levinson, H.V.   Barron, D. Botstein, C. Kenyon (descubridora en nematodos, de  genes que alargan   sus vidas), R.L. Cohen y JW Lewis,  trabajan con este fin.  Conjuntamente con la   compañía AbbVie, Calico, planea invertir $1.5 billones en investigaciones y elaboración de medicamentos  anti-envejecimiento.   Otra  compañía:  Human Longeviy, liderada por Craig Venter, centra sus estudios directamente en humanos, esperando secuenciar  los  genes de 100,000 personas  por año, para analizar sus perfiles proteicos, contenido microbiano de sus cuerpos y digitalización de  imágenes de sus somas.   Algunas drogas descubiertas son los compuestos P7C3, el resveratrol, la metformina, sirtuinas.
II) Para nosotros el éxito de estas compañías debe basarse  en  concepciones holísticas actualizadas del sistema  crecimiento-envejecimiento en  humanos. Veamos: a) Sabemos que los primeros mamíferos (de quienes descendemos),  evolucionaron de reptiles, quienes a diferencia de los primeros,  no incrementan su mortalidad con la edad, regeneran oocitos y  miembros y reemplazan continuamente sus dientes, hipotetizandose que a causa del tamaño pequeño de  los  mamíferos primigenios, estos  adoptaron la  estrategia evolutiva de sobrevivir y  reproducirse tempranamente, para luego envejecer rápidamente, con lo que la mayor  contribución de la selección natural para engendrar  nuevas generaciones, provendría  fundamentalmente de los jóvenes. Por lo expuesto y según Peter Medawar (1952), la fuerza positiva de la selección  disminuye con la edad. El reciente descubrimiento de la proteína  GDF11, en la sangre de ratones  menores de 3 meses de edad, parece  confirmar esta teoría.  Según el modelo de  William, algunos genes benéficos durante la infancia se transformarían en peligrosos (acumulación de mutaciones),    durante el envejecimiento.  b)  Kirkwood  y  Austad  (2000), sostienen que el  envejecimiento ocurre por  acumulación de daño celular  y  el desgaste energético inducido  por  múltiples mecanismos defensivo-reparativos,   empleados a lo largo de nuestras vidas. c) Las compañías mencionadas y otras, desarrollan   a este fin,  medicinas especificas (targeted medicines), usualmente creadas mediante  ingeniería  genética.  La elaboración de estos  medicamentos  específicos esta focalizada por el momento en  las  enfermedades más prevalentes en personas que envejecen: cáncer, diabetes  y desordenes neurodegenerativos.  d) Como se ve las compañías  biofarmaceuticas  mencionadas      no van a investigar  ni tratar  el problema de fondo del envejecimiento,  el que según  L. Hayflick, ocurre tras  alcanzar la madurez reproductiva, cuando se inicia una  inestabilidad termodinámica, que descompone la maquinaria somática (genoma), que mantenía  el orden y  eficacia  molecular (reemplazo y reparaciones celulares adecuadas), llevando a Hayflick a realizar la pregunta fundamental en biología del envejecimiento ¿qué cambios en las  biomoleculas llevan a un incremento de la vulnerabilidad y declinación funcional del organismo?.e) Una ingeniosa solución anti-envejecimiento es la sugerida   por Haldane: posponer la maduración sexual, con medicamentos tanto  como  sea posible (a los  120 años, tendríamos  cuerpos de niños).

ANTI-AGING EFFECT  in ADULT MICE

The recent discovery of anti-aging effects in adult mice after blood transfusions coming from young mice  -from our point of view-  is solid and  have  human perspective, because: a) One of five,   identified antiaging protein : growth differentiation factor 11 (GDF11), in the serum  of  young  donor mice is similar to that in humans. b) The physiology of young and senile mice is different. In young mice prevail growth promoting factors : in size and volume, as well as functional efficiency of cells (synaptic plasticity, cognitive efficiency) and better organs (best bodies). Conversely, in aged mice, the body tries to avoid wear and functional decline of the brain, heart, muscle strength, etc.). c) It would be interesting to investigate whether blood transfusion of old mice to young mice, slows growth and development of the last.

EFECTO  ANTIENVEJECIMIENTO,  en   RATONES  ADULTOS


El reciente descubrimiento  de efectos antienvejecimientoen ratones adultos,  tras transfundirles  sangre  procedente de ratones jóvenes, tiene según nuestro punto de vista solidez  y perspectivas humanas, porque: a) Una de las 5 proteínas antienvejecimiento  identificadas :  growth differentiation factor 11 (GDF11)en el suero  de los ratones jóvenes donantes,  es similar a la existente en humanos. b) La fisiología de ratones jóvenes y seniles, es distinta.  En ratones jóvenes priman  factores promotores del crecimiento en  tamaño y  volumen, así como eficacia funcional  de  células (plasticidad sináptica, eficacia cognitiva) y órganos (mejores cuerpos).  Contrariamente, en ratones de edad avanzada,  prima  evitar el desgaste  y la  declinación funcional del  cerebro,  corazón, de la fuerza muscular, etc.).c) Sería interesante  investigar si la trasfusión de sangre de ratones  viejos a ratones jóvenes, enlentece  el crecimiento y desarrollo, de estos.

LAUNCH  of   HINDU ROCKET: IMPLICATIONS
 
The recent launching  (16/12/2014), of the  Geostationary Satellite Launch Vehicle (GSLV), Mk- III, rocket by the space agency : ISRO, places  Hindus to the level of  : Chinese, American European and Russian  space agencies, because: I) The GSLV is a rocket of 630 tons, for terrestrial and extraterrestrial long range missions (interplanetary and multi-satellite launches).  Let's look its  unmanned capsule (designed to accommodate 3 human crew),  of 3735 kg , which was separated and landed successfully in the Indian ocean, 20 minutes after departure, destined to descend on the Moon by 2017 and by 2018, on Mars. II) The GSLV employs a powerful cryogenic engine, continuously upgraded. III) Show the world the existence of a successful upscale techno-scientific Hindu knowledge. IV) The synthesis of this human group  focused on priorities (without neglecting the practical), achieve successful launches with limited financial budgets. The cost of the recent launch of GSVL, was only $ 25 million. V) To make matters better, India is beginning to reap profits after entering to a market of $ 300 billion, advising and assisting developed countries like:  Germany, France, Canada, etc in space and related fields. 3 years ago,  Hindus discovered in the upper atmosphere (height : 27-30 km), 3 species of extremophile bacteria (one called: Bacillus isronensis), extremely resistant to ultraviolet radiation. 

IMPLICANCIAS del  LANZAMIENTO DE COHETE HINDU


El reciente lanzamiento (16/12/2014), del cohete  Geostationary Satellite Launch Vehicle (GSLV), Mk-III, por parte de la agencia espacial: ISRO,  sitúa a los hindúes al mismo nivel  que las agencias espaciales: americana,  china,  europea y rusa por: I) El GSLV, es un cohete de  630 toneladas, para realizar misiones terrestres y extraterrestres de largo alcance (lanzamientos interplanetarios  y   multisatelitales en orbitas altas). Fijémonos nomas en la capsula no tripulada (destinada a albergar 3 tripulantes), de  3735 kg de peso, que se separo y aterrizo con éxito en el mar hindú,  20 minutos  después de la partida, destinada a  descender  el   2017 en la Luna y el 2018, en   Marte. II) El GSLV, emplea un poderoso motor  criogénico, en continuo perfeccionamiento. III) Muestra al mundo la existencia de un saber tecno-científico hindú exitoso y de  primer nivel.  IV) La capacidad de síntesis de este grupo humano, esta focalizada en  prioridades (sin descuidar lo práctico), logrando exitosos lanzamientos espaciales con presupuestos económicos reducidos.   El coste del reciente lanzamiento  del GSVL, fue de  apenas   25 millones de dólares. V) Por si fuera poco, India empieza a cosechar ganancias al ingresar a un mercado de 300 billones de dólares, asesorando y asistiendo a  países desarrollados como Alemania, Francia, Canadá, etc  en materia espacial y relacionados.  Años atrás los hindúes,  descubrieron en la atmosfera superior (altura:  27–30 km), 3 especies de   bacterias extremofilas (una de ellas : Bacillus isronensis),   extremadamente resistentes a la radiación ultravioleta.

OPTOGENETICS

Thinking  about   the difficulties and slow progress in brain research, Crick (1999. 354: 221-25), wrote: "a)...When we  understand scientifically our perceptions, thoughts and emotions, the vision of ourselves and the universe will be totally transformed ...b) ... After identifying the different types  of neurons, the next big step will be  to put one or more neurons quickly, in on-off, using infrared light ... in a particular light sensitive cell”. If we wanted to scrutinize today, one of the avenues through which will pass  techno-science very fast in the coming years, the selected would be, no doubt : optogenetics. Some of   leading researchers in this area: Karl Deisseroth, Edward S. Boyden, Feng Zhang, Peter Hegemann, Gero Miesenböck, Georg Nagel, etc., may soon receive a Nobel Prize. Boyden (MIT), says that his  optogenetic investigations are possible because the brain (organ, about which many  thought we would never understand its operation), works  with binary codes that can be explored and understood, easily. If so, it would be possible for us to be bio-robots with binary codes programmed by nature or someone. Therefore,  artificial intelligence with programmable codes and emerging roles  should reach the same position and sophistication of modern human brains.

Current experiments in optogenetics, excite (on, gain of function) or inhibit (off, loss of function), brain cells of experimental animals in vivo, shooting rays of light rays  to  neuronal surfaces, previously sensitized with rhodopsin (protein sensitive to visible light), extracted  from environmental microorganisms, allowing researchers  to perform accurate temporary registers with optogenetic sensors for calcium, chlorides, voltages membranes, etc. Preconditioning of neurons is performed  translating opsin-genes (via  virus carriers), extracted from  envirommental, unicellular  fluorescent algae to  neurons of   experimental animals. Using channelrhodopsins to excite  neurons or silencing them with halorhodopsins,  allow us  a  better understanding about how normal and abnormal neural circuits, functions. It is  expected soon to know the causes of certain neurological diseases (Parkinson disease, autism, schizophrenia, drug addiction, anxiety, depression, etc.), to treat them properly. It is also expected to control inappropriate behavior of animals and why not of certain humans.

OPTOGENETICA

Reflexionando en torno a las   dificultades  y  lento  avance   de las investigaciones cerebrales, Crick (1999. 354:221-25), escribió: “a)Cuando comprendamos científicamente  nuestras percepciones, pensamientos y emociones, la visión de nosotros mismos y del universo será totalmente transformada... b)Tras identificar las diferentes clases  de neuronas, el  siguiente  gran paso será poner rápidamente,  una o varias neuronas  en on-off, usando luz infrarroja…en una   célula particular,  sensible  a la luz”. Si deseáramos  escudriñar hoy, alguna de las  avenidas por las que discurrirá aceleradamente  la  tecno-ciencia  en los próximos años, la seleccionada seria a no dudarlo, la  optogenética. Alguno de los investigadores  destacados  en  esta área: Karl Deisseroth, Edward S. Boyden, Feng Zhang, Peter Hegemann, Gero Miesenböck, Georg Nagel, etc., podría recibir pronto un    Premio Nobel. Boyden (MIT), sostiene que sus investigaciones  optogenéticas son posibles porque el  cerebro (órgano, del  que se pensaba que nunca entenderíamos  su funcionamiento),  trabaja  con códigos binarios, susceptibles de ser explorados y comprendidos, fácilmente.  De ser así, existiría la posibilidad de que seamos  bio-robots con códigos binarios programados por  la naturaleza o alguien.  Por tanto, la inteligencia artificial con códigos programables y funciones emergentes debería  alcanzar  la misma tesitura y sofisticación de los cerebros  humanos actuales.

Los experimentos optogeneticos actuales,  excitan (on, gain of function) o,  inhiben  (off, loss of function), neuronas cerebrales de animales de experimentación  in vivo, lanzando rayos de luz a  superficies  neuronales   acondicionadas  y sensibilizadas previamente  con   rodopsinas (proteínas sensibles a la  luz  visible), procedentes de microorganismos ambientales, permitiendo realizar registros temporales precisos con sensores optogeneticos  para el calcio,cloruros,voltajes de membranas,  etc. El acondicionamiento previo de las neuronas se realiza transportando genes-opsina (con virus),  extraídos de algas unicelulares fluorescentes ambientales a neuronas de animales de experimentación. excitando neuronas con channelrhodopsins o silenciándolas con halorhodopsins,  permitiendo  comprender mejor el funcionamiento de  circuitos neurales normales y patológicos, esperándose    conocer pronto, las causas de ciertas enfermedades neurológicas (E. de Parkinson, autismo, esquizofrenia,  adicción a drogas, ansiedad, depresión, etc.), para  tratarlas adecuadamente. Se espera asimismo controlar conductas inapropiadas de animales y porque no, de  ciertos  humanos.
