DESIRED GENES




GENE DRIVES


If CRISPR technique is a molecular biological genomic text processor, that can alter genetic information of any organism; its derivative:  Gene Drives application, seems to have an extraordinary potential to eradicate entire biological species, be a biological weapon  and/or control certain diseases, optimize agriculture, stop the spread of malaria, manage wild ecosystems, etc., introducing desired genetic traits in animals, plants and even human populations. The point is no longer about having programmed children with blue eyes, but to create for example: animals free from all kinds of infectious diseases and so on.  1-The desired gene drive-CRISPR, is inserted into the chromosome of an organism that is sexually mating. 2- The inserted gene drive-CRISPR cleaves a specific portion of the corresponding chromosome from the other parent. 3- Copies of the gene drive-CRISPR of the first parent are made. 4-Then, autoduplicated gene drive-CRISPR restores empty (cut), spaces on both chromosomes, happening the same in each original descendants of the body, ensuring 100% of gene drive transmission to subsequent generations. Although this technique works well on a small scale in fruit flies, mosquitoes and yeasts, it is to be demonstrated that  the introduced desired trait is able to defy the rules of inheritance, in particular: the evolutionary resistance, that will emerge especially in large populations. In order that   gene drive technique accomplish its mission, it is necessary that transmission to the next generations be 100% and not : 50-50% that happened in ordinary conditions. It is argued that once introduced a gene drive it is impossible to reverse ecological damage (if it happened). According to Philipp Messer (Cornell University), only the emergence of natural resistance -after several generations- is the only possibility of eliminating the harmful action of a gene drive. Gene drive application works by replacing (alteration of a specific trait), by which for example a gene drive is able to change the genome of a mosquito, disabling it to carry the malaria parasite. And, by deletion, that is for example: a gene drive would be able to force the offspring of a mosquito to disable all its male reproduction. According to Flaminia Catteruccia (Harvard University), wild populations are able to develop resistance to gene drives, using the option: nonhomologous end-joining rate (cut genomic ends are repaired at random). That is, if the nonhomologous end joining are unchecked, the organism inevitably will develop resistance to scale, initiated somewhere in the genome, which will   spread later, to the surrounding environment. Thus, malaria parasite would be resistant, mutating into a new infectious form in a mosquito previously made resistant by genetic engineering. Lately, Messer has tracing drives gene action in   5000 fruit flies marked with a red fluorescent protein indicator of how far the gene drive action came until the emergence of resistance. Multiple genes are also inserted -instead of a single gene- to create immune   resistance mosquitoes

GENE   DRIVES


Si la técnica   CRISPR, es un procesador molecular  de  textos biológico-genómico- capaz de  alterar la información genética de cualquier organismo, su derivado  : la aplicación Gene Drives, parece tener   la extraordinaria potencialidad de erradicar especies biológicas enteras, ser un arma biológica  y/o,  controlar ciertas enfermedades, optimizar la  agricultura, detener la diseminación de la malaria, manejar ecosistemas salvajes, anular la resistencia a antibióticos, reprogramar genes de mosquitos,etc.,   introduciendo  rasgos genéticos en poblaciones   animales,  vegetales y hasta  de seres humanos. Ya no se trata de tener hijos programados con ojos azules, sino de crear poblaciones animales enteras, libres de todo tipo de enfermedades infecciosas, por ejemplo.  Para ello: 1- Se inserta el  gene drive-CRISPR, en un sitio específico del cromosoma de un organismo que se está apareando  sexualmente.  2-El gene drive CRISPR introducido corta una porción especifica del cromosoma  correspondiente del  otro padre. 3- Se  realizan copias del gen drive del primer padre conteniendo la sección: gene drive-CRISPR. 4-El gene drive  autoduplicado  restaura  los espacios genómicos vacíos (cortados), en ambos cromosomas, asegurando una trasmisión 100% del gen drive a las siguientes generaciones. Aunque esta técnica funciona bien a pequeña escala en la mosca de la fruta, mosquitos y levaduras, el asunto a demostrar es, si el  rasgo deseado introducido  es capaz de desafiar las reglas de la herencia, en especial: la resistencia evolutiva, presta a surgir en grandes poblaciones. Para que la técnica gene drive cumpla su cometido, se necesita  que la  trasmisión  a las siguientes generaciones sea 100% y hasta ahora un rasgo especifico en condiciones ordinarias, solo tiene una chance: 50-50 %, de ser trasmitido. Y, se  aduce que  una vez introducido un gene drive es imposible revertir un daño ecológico (si ocurriese), siendo según Philipp Messer (Universidad de Cornell),  solo la emergencia de  la resistencia  natural -después de varias generaciones- la única posibilidad de eliminar  la acción nociva de un gen drive.  La aplicación gen drive, funciona mediante reemplazo (alteración de un rasgo específico), mediante el cual un gen drive es capaz de  cambiar el genoma de un mosquito, inhabilitándolo por ejemplo a portar el parasito de la malaria. Y,  mediante supresión en la que un gene drive -por ejemplo- sería  capaz de forzar  a la descendencia de un mosquito a inhabilitar la totalidad de su  reproducción masculina.  Según Flaminia Catteruccia (Universidad de Harvard), las poblaciones salvajes son capaces de desarrollar resistencia a  los genes drives, mediante la opción: nonhomologous end-joining rate (extremos genómicos cortados se reparan al azar). Es decir,  si no se  controla  el  nonhomologous end joining, se desarrollara inevitablemente  resistencia a escala, iniciada en algún punto del genoma, que luego se extenderá  al medio ambiente circundante. De este modo, el parasito de la malaria se haría  resistente, mutando  hacia  una nueva forma infecciosa en un mosquito hecho previamente resistente a la malaria mediante ingeniería genética. Últimamente, Messer, ha  rastreando la acción  de gene drives en las cajas de 5000 moscas de la fruta señalizada  con  una proteína fluorescente de color rojo   indicadora de  cuán lejos llego  la acción de los gene drives hasta la emergencia   de  la resistencia. También se  insertan múltiples genes -en lugar de un solo gene drive- para  crear mosquitos   inmunes a la resistencia

JUPITER


JUPITER, THE POTENTIAL  DARK  SUN
In the last four days we analyzed  the photos  of   Jupiter’ surface, its poles and some of its satellites, sent to earth by the Juno probe. In this regard, it seems important to comment on two aspects:  I) The images sent are corroborative, but modestly novel, not changing much  the   overview and known  aspects  on this planet. Just a little more than  the  inside  provided by  images sent by the   Cassini-Huygens  probe  (2004), as it passed   by Saturn,  ratifying   Jupiter as a gigantic and rugged gaseous planet,  although  it  provided   little information about  its  internal functional structure. While the international press talks now about   completely different photos, the  real  thing  is that Jupiter is obviously different. Jupiter is not a rocky but a gaseous planet (and therefore different). That’s why Jupiter  will  continue  being so   different.  Nevertheless there are some  solid data  missing  from its rocky inner core,   the significance    of the great red spot, the movement of his innermost layers, its high speed of rotation on its axis and others. Certainly,  their north and south poles not look white as in  Earth, but blue for reasons that are already being explained. These,  does not change things so much. The photos sent by Juno  confirm what was already known: a rather turbulent and bleak picture. Today, Jupiter is an  uninhabitable planet. But if we tried to find something good to earth human beings in  this pessimistic outlook, we may find something  at  long term. II) To answer the above may be necessary to ask: Why we must  explore this planet? What benefits can bring this kind of research? Although paradoxical, the matter is that if we confirm certain assumptions about the consequences of   gas structure of this planet, maybe after a few thousand years (seconds in the history of the cosmos), it will allow earthlings live in a future  not only on Jupiter, but also in one of its satellites, because Jupiter is almost a second sun. An emergent  potential dark sun, when the current sun will become a white dwarf that will disappear along with all   rocky planets (Earth, Mars, Mercury). When that happens Jupiter and  Europa  (with its vast oceans),  will remain in its orbit, given its independence from the sun of our planetary system. Regarding the issue of protecting our planet, Jupiter has its own  history.  4000 million years ago  Jupiter swallowed  the ocean of Mars, during a migration out of its orbit (performed also by Saturn), respecting the oceans of  Earth. Jupiter  has also served as a protective shield of our planet avoiding the impact of comets and asteroids on our planet that could have  eliminated  all forms of human life. That is the reason to study this giant.
JUPITER,  EL  POTENCIAL  SOL OSCURO
En los últimos 4 días analizamos  las fotos de la superficie de Júpiter, sus polos  y  algunos de sus satélites, enviadas a la tierra por la sonda Juno. Al respecto,   nos parece importante comentar 2 aspectos: I) Las  imágenes enviadas son confirmativas, pero modestamente novedosas, no  cambiando  mucho la visión general   ya conocida de este planeta. Apenas un poco más del inside  proporcionado  por  las imágenes enviadas  por la  nave-sonda  Cassini-Huygens (2004), a su  paso por  Saturno  que ratifico a  Júpiter como un planeta gaseoso gigantesco  y agreste aunque proporciono  poca información de su aspecto  funcional interno. Aunque la prensa internacional habla ahora, de fotos totalmente diferentes, lo real es que el Júpiter  diferente es obvio. Júpiter  no es un planeta rocoso  sino   gaseoso (y por lo tanto diferente). Es decir,  continuaremos viendo cosas  diferentes de él.  Empero,   faltan obtener  datos más sólidos de    su núcleo central  interno rocoso, de la razón de ser  de su gran mancha roja, del  movimiento de sus capas más internas, de su gran velocidad de rotación sobre su eje y,  otros.   Que sus   polos norte   y sur  no luzcan   blancos   como los de la  Tierra,  sino  azules  por razones que  ya están siendo  explicadas no cambia mucho las cosas. Las fotos enviadas por Juno   confirman lo ya conocido : un panorama  bastante turbulento y desolador.  Hoy por hoy,   un planeta inhabitable. Empero si tratásemos de encontrar algo bueno para los terrícolas dentro de esta perspectiva pesimista,  quizás descubramos   algo a largo plazo. II) Para responder a lo  anterior quizás sea necesario  preguntarse: ¿Para  qué explorar este planeta? ¿Qué beneficios puede traernos  este tipo de investigaciones?  Aunque paradójico,  el asunto es que el confirmar  ciertas  presunciones sobre las consecuencias de la estructura gaseosa de este planeta, puede   que   después de algunos miles de años (segundos en la historia del cosmos), permita a los terrícolas  habitar   no solo en Júpiter,  sino también en  alguno de sus satélites, porque Júpiter es casi un segundo sol. Un potencial  sol  oscuro, cuando el sol actual se convierta en una enana blanca que desaparecerá conjuntamente con  todos los planetas rocosos (Tierra, Marte, Mercurio). Cuando eso suceda Júpiter y Europa (con sus inmensos océanos), permanecerán en su órbita, dada su independencia del sol actual de nuestro sistema planetario. Respecto al tema protector de Júpiter sobre nuestro planeta hay antecedentes.  Hace   mas o menos  4000 millones de años Júpiter se tragó el océano  de Marte durante una migración fuera de su órbita (que también realizo Saturno), respetando los   océanos de la Tierra. Asimismo,  ha servido de escudo protector de nuestro planeta  evitando el impacto de  cometas y asteroides sobre nuestro planeta que de otro lado hubiesen podido eliminar toda forma de vida humana.  Esa es la razón para  estudiar a este gigante.
