ARTIFICIAL WOMBS





NOAH’S ARK, 2030.  If a major world catastrophe occurred: nuclear hecatomb or impact of a large asteroid on Earth and only survive in powerful bunkers a very small portion of humans with the best remaining technology. How would repopulate this minority the entire world? In Brave New World (1932), Aldous Huxley conceived enormous canvases hung  on the walls, serving  as artificial wombs for total extra-uterine gestations. Altough this fiction still  does not  materialize, we begin to approach it. 6 preterm fetuses of  lambs equivalent  to 23-week-old human fetuses were able to complete their development  in artificial wombs  for the first time  :transparent  biobags  filled with amniotic fluid substitutes connected to gas exchangers  attached  to  the umbilical  cords of floating lambs,  providing them with oxygen, nutrients and chemicals, to stimulate their  growth.  Biobags  built at a hospital in Philadephia (Nature Communications).

Si ocurriese una  gran catástrofe mundial : hecatombe    nuclear o  impacto de un gran asteroide sobre  la Tierra y solo sobreviviesen en poderosos bunkers   una pequeñísima  porción de humanos  con la  mejor tecnología remanente ¿Cómo haría esta minoría para repoblar el mundo?. En Brave New World (1932), Aldous Huxley concibió  enormes lienzos colgados en las  paredes fungiendo de úteros  artificiales para gestaciones extra-uterinas totales.   Aunque esta ficción aún no se materializa, nos empezamos a acercar a ella.  6  fetos prematuros  de corderos   cesareados  equivalentes a  fetos  humanos de   23 semanas lograron  por primera vez completar  su desarrollo   en úteros  artificiales : biobags trasparentes, llenos de sustitutos de liquido amniótico conectados a intercambiadores de gases unidos a los  cordones umbilicales de corderos  flotantes,  proveyéndoles de oxígeno, nutrientes y químicos para estimular  sus  crecimientos.  Biobags construidos en un hospital de Filadelfia (Nature Communications)

CHROMATIN REMODELING

There are some important questions about DNA: how and where was DNA formed, for the first time: on Earth? In outer space?. What laws make possible its stability and sustained self-reproduction? For us, DNA obeys the same laws that govern the natural world and therefore -over time- we will be able to create artificial DNAs not only with the same functions, but improved  (longer useful life, improved genes to prevent and fight many diseases, including cancer, that our cells generate more ATP,  stronger bones and teeth, skin automatically adaptable to climatic changes, etc.). To achieve this goal is necessary to arrive at a holistic understanding of DNA  functioning, with the help of science and technology. With respect to the latter, the rapid and increasing understanding of  cellular packing and repair mechanisms of the cellular chromatin with the use of the cryo electron microscopy (Cryo EM), which freezes the biological samples, cuts, aligns and  showed   them   in 3D, allows to confirm hypothesis and to create others, at great speed. A recent paper by Xiao Li et al  published in Nature, explains the organization of massive genomes in order to remain functionally stable and the mechanisms of packaging and remodeling of chromatin, revealed by the paired structure of nucleosomes-Snf2 enzymes, the latter capable to alter the structure of the chromatin and positions of cellular nucleosomes, allowing   regulatory proteins to access to the DNA. The Cryo  EM allowed to visualize the  ​​union of the 2 main domains of these enzymes to the nucleosome to activate, to allow  coupling and  ATP  hydrolysis, for the remodeling of the chromatin. The enzyme Snf2, binds to the phosphate pillars of a DNA gyre  (nucleosome), using mechanisms employed  with different remodelers. Finally the enzyme deforms locally the nucleosome of the DNA in the area of ​​union initiating the reaction of remodeling. Although the work did not detect the protein that function as an engine (perhaps:  cohesin protein complexes or RNA polymerases), to allow the extrusion of DNA loops, the study only discovered already known mechanical and topological mechanisms. An important work because the extrusion of DNA loops, holds together many local regions of the genome, protected by a central organizing principle, which prevents the entanglement   of the loops while   chromosomes are formed. Although the authors believe that studying the proper conformation of DNA loops or their deviations will serve to detect health and cancer states, the energetic mechanism that forms these loops continues to be a mystery.

De las variadas preguntas existentes, respecto al DNA hay  algunas  importantes como: donde y como se formo el DNA,por primera vez: en la Tierra? En el espacio exterior?.  Que leyes hacen  posible su estabilidad y sostenida  autoreproduccion?. Para nosotros,  el DNA obedece a las mismas leyes   que  gobiernan  al mundo  natural y, por  lo tanto -con el tiempo- seremos capaces de   crear  DNAs artificiales  no solo con  las mismas funciones, sino  mejoradas (vida util mas prolongada, genes mejorados para prevenir y combatir muchas enfermedades, incluido el cancer,  que nuestras celulas generen mas ATP,  que  generen huesos y dientes mas fuertes,  piel automaticamente adaptable a   cambios climaticos,etc). Para lograr esta meta es necesario arribar a  una comprension holistica del funcionamiento del DNA,con la ayuda de la ciencia y tecnologia. Respecto a esto ultimo, se avanza rapidamente, siendo  prueba de ello  la  acelerada y creciente comprension de parte de  los mecanismos de   empaquetamiento y reparacion de la cromatina  celular  mediante el empleo del  microscopio electronico en frio (MEF), que congela minusculas muestras biologicas, las corta, las  alinea y tras  visibilizarlas  en  3D, permite  confirmar  hipotesis y crear  otras, a gran velocidad.  Un reciente trabajo de Xiao Li y colaboradores publicado en Nature,   explica la organizacion de genomas masivos a fin de  mantenerse funcionalmente estables y los mecanismos de empaquetamiento y remodelacion  de la  cromatina, revelados por la estructura apareada de  nucleosomas-enzimas  Snf2,  estas ultimas Year published:capaces de alterar  la  estructura de la cromatina  y las posiciones de  nucleosomas celulares,  permitiendo a las proteinas regulatorias acceder al DNA. El MEF permitio visualizar la union de los 2 principales dominios  de  estas enzimas  al  nucleosoma  para  activarse,  permitir  el acoplamiento e  hidrolisis del  ATP, para la remodelacion de la cromatina.  La enzima Snf2, se une a los   pilares del fosfato de un giro del  DNA (nucleosoma), mediante   mecanismos empleados con diferentes remodeladores. Finalmente la enzima deforma  localmente al nucleosoma del  DNA en el area de union  iniciandose la reaccion de remodelacion. Aunque el trabajo no detecto a la proteina que hace de motor (se sospecha de complejos proteicos de  coesina y  RNA polimerasas), para permitir la extrusion de asas de DNA, el estudio solo descubrio mecanismos mecanicos y topologicos,  ya conocidos. Un  trabajo importante porque  la  extrusion   de asas de DNA,  mantiene unidas  muchas  regiones locales del  genoma, al amparo de un principio organizador central, que impide el atascamiento  de las asas mientras se  forman  los cromosomas.  Aunque los autores opinan que el estudio de la conformacion adecuada de las asas de DNA o sus desviaciones  serviran para detectar estados de salud y cancer, el mecanismo energetico que forma estas asas continua siendo    un misterio.
